Our goal is to understand the role of ubiquitin-like molecules (Ubls) in transcriptional activity regulation and cell growth. In particular, we are focusing on the identification and characterisation of novel targets for the NEDD8 Ubl and how the NEDD8 pathway is controlled in cells. We develop a multi-disciplinary research programme, combining biochemical and biological methods with innovative quantitative (SILAC) proteomic and microscopy approaches in human cells and animal model systems (C. elegans). We identified the p53 tumour suppressor and the Mdm2 E3-ligase as the first non-cullins substrates for NEDD8. Through our proteomic approaches we then expanded our studies on protein NEDDylation and reported the first NEDD8 proteome in mammalian cells. Following on the proteomic studies we characterised a new role for NEDD8 in the function of the nucleolus as a sensor of perturbations in cell growth. Additional studies discovered roles for NEDD8 in protein trafficking, and how apoptotic signals and chemotherapeutic drugs can elicit their effects through regulation of NEDDylation in cells. We recently discovered that the NEDD8 pathway is a sensor of a variety of stress signals, including proteasome inhibition, heat shock and oxidative stress. Our goal is to understand the molecular basis and biological implications of the NEDD8 response to cellular stress.
